Glucose oxidation and cytochromes in solubilized particulate fractions of Acetobacter suboxydans.

چکیده
Microbial terminal oxidases are usually associated with particulate fractions in the cell. These fractions, however, are difficult to resolve into smaller active molecules that are suitable for enzymatic studies in vitro. This fact led Rittenberg (1) to propose that these particles are surrounded by a special membrane. The present paper will report a method of “solubilization”’ of particulate oxidase systems from Acetobacter suboxydans. This method can also be applied to other organisms, such a.s Acetobacter pasteurianum. With the extracts obtained, cytochrome spectra and a glucose oxidase have been observed.

Acetobacter suboxydans is known to bring about oxidation of polyhydroxy compounds. The action of the bacterial enzyme is limited, however, to substances with specific configurations. According to the generalization of Bertrand,’ only those hydroxyl groups are oxidized which are situated between a primary hydroxyl group and a secondary one in a cis position. متن کاملGlucose and gluconate dissimilation in Acetobacter suboxydans.
Investigations into the nature of the metabolism of glucose in Acetobacter suboxydans have revealed that both the pentose cycle (1) and a direct nonphosphorylative oxidation (2) contribute to the consumption of this substrate. Fructose, in like manner, dissimilates by way of the pentose pathway (3) as does glycerol, after its conversion to fructose-l ,6-diphosphate via triose phosphate (1,4). S...
متن کاملTricarboxylic acid cycle activity in Acetobacter pasteurianum.
Previous papers from this laboratory (King and Cheldelin, 1952, 1952a, 1954; Hauge et al., 1955, 1955a) have reported on the oxidation of carbohydrates in Acetobacter suboxydans. Neither resting nor disintegrated cells can oxidize acetate or other intermediates of the tricarboxylic acid cycle. These findings have recently been confirmed in another laboratory (Rao and Gunsalus, 1955). From the c...
متن کاملSources of energy and the dinitrophenol effect in the growth of Acetobacter suboxydans.
In resting Acetobacter suboxydans cells, glycerol can be oxidized beyond the dihydroxyacetone stage, and sorbitol, beyond the sorbose stage (King and Cheldelin, 1952a). 2 ,4-Dinitrophenol inhibits the further oxidation of dihydroxyacetone or sorbose but has no effect upon the first step oxidations (King and Cheldelin, 1952b). Since these keto derivatives can be metabolized further by resting ce...
